什么是连续流动化学?

连续流动化学在化学和石油化学行业里已有几十年的应用。这种连续流动化学的生产手段正在制药研发中受到重视,考虑到其以下优势:

  • 更好的工艺过程安全性
  • 更优的质量
  • 空间的节省
  • 更高的产能

以其最简单的形式,连续流动化学始于两种以上的物料 — 比如起始反应物。这些物料流以设定流速用泵打入一反应舱室、反应管、有时是一个具有使流动物料进行混合和反应的数多小槽的微型反应器。流进反应舱室的不同反应物料在此进行混合和反应。由于这些微型反应器的舱室或管的体积非常小,仅需小量的物料进行反应,因此:

  • 减少昂贵试剂/化合物的用量
  • 降低溶剂消耗
  • 改善温度控制
  • 提高工作安全性(由于潜在爆炸性物料量的降低)。

根据反应动力学和物料流速,需要保证反应物料在微型反应器中达到某一特定的停留时间,从而获得预期的反应转换率。相继,从微型反应器出口流出的物料用烧瓶或其它适当的容器收集起来。

因为反应是在连续流动的流体中进行,自然希望对反应进行监测以便得知各种反应条件状况,包括:

  • 稳定状态
  • 扩散特性
  • 反应中间体的存在

对连续流动反应的监测要求监测技术能在流动的流体中分辨不同反应组分。与微型流动池相匹配的反应红外仪(ReactIR)是一适宜的技术。ReactIR基于FTIR光谱并结合(变角)衰减全反射(ATR),是用于原位测量和跟踪反应组分的理想工具。

FTIR(傅立叶变换红外)光谱分析对某一物料的特征官能团给出特定的指纹图谱。它为识别和鉴定反应组分提供了便利。通过在一定时间里测量相应的IR信号强度,便能跟踪反应期间各种组分浓度的相对或绝对值。换句话说,你能看到反应趋势的变化,并且当趋势平稳不变时你可以判断反应达到稳定态。
